Histological and Immunohistochemical Study of Endocrine Pancreas in Common Quail (Coturnix coturnix)

Abstract
The purpose of this study is to characterize the histological and immunohistochemical features of endocrine
pancreas of common quail (Coturnixcoturnix).Small pieces of tissue from ten apparently healthy adult quail
males pancreaseswere dissected after slaughtered. Sections were stained with Gomorra’s method stain for
distinguish pancreatic islets .Sections were also processed for standard immunohistochemistry techniques
using polyclonal primary and secondary antibodies for glucagon and insulin hormones .The histological
examination showed three types were identified: alphais lets were mainly composed of alpha and delta cells,
beta islets were mainly composed of beta and delta cells and mixed is lets were mainly composed of alpha,
beta and delta cells. Furthermore alpha cells were round, oval or columnar in shape, beta cells appeared as
round, elliptical or sometimes appeared polygonal in shape, delta cells are small in size, roughly spherical or
polygonal in shape.Immunohistochemical staining of endocrine pancreas revealed glucagon immune reactive
cells occupying most of the dark islets, periphery of mixed islets whereas insulin immune reactive cells in the
center of beta islets and mixed islets. In conclusion, the endocrine histology and immunohistochemistry
technique results of quails are similar to observations in other bird species such as chickens, ducks and geese.
