Preparation, Characterization and Cytotoxic Evaluation of Paclitaxel loaded PLGA Nanoparticles Using Novel Polymer Soluplus® as Surfactant

Abstract
This study aims to prepare polymeric nanoparticles using PLGA. A double emulsion solvent evaporation method
as well as a single emulsion solvent evaporation method were used to formulate the nanoparticles using Soluplus®
as the surfactant. In order to encapsulate the hydrophobic drug Paclitaxel, a variety of formulations were designed
and tested in terms of particle size, polydispersity index, zeta potential, drug loading, and entrapment efficiency.
Morphology was observed using scanning electron microscopy. MTT assay was used to determine whether
nanoparticles are more cytotoxic than free drugs. It was shown that nanoparticles performed better than free drugs
in terms of cytotoxicity as a result of successful fabrication of the nanoparticles.
